From Montevideo, you can take a bus or a car via Route 1 to Carmelo which takes about 3 hours. From Carmelo, you can take a bus or a car via Route 21 and Route 10 to Punta del Este which takes about 4-5 hours.

All the three places are coastal cities and are pretty much at sea level, so you needn’t worry about altitude sickness.
